Summary

Violation of the native conformation of a protein molecule not only denatures it and deprives it of the ability to perform its biological functions, but also destabilizes its structure. Under normal conditions, this leads to the rapid removal of the molecule from circulation. At insuffiency of clearance systems the accumulation of destabilized molecules is observed. Such protein derivatives show a tendency to aggregation. A typical example of such processes is the case of diabetic foot – a common complication of diabetes mellitus, in which progressive tissue degeneration occurs. The aim of this work is the study of the features of structures formed at early and more advanced stages of tissue degradation. It is shown that at early stages of tissue decay, protein spherulites are formed. These structures are formed by β-sheet structures that grow from a starting point of nucleation. For later stages of tissue decay, deposits of amyloid-like aggregates are formed. The molecular mechanisms of these pathological processes are considered.
